Product Features and Benefits: Amine reactive AND carbonyl/carboxyl reactive pegylation crosslinking reagent Unique dPEG containing HETEROBIFUNCTIONAL crosslinker with amine (NHS) and carbonyl reactive (protected-hydrazide; which can be deprotected using TFA or HCl) The hydrazone formed from the incipient hydrazide can be used as a cleavable linking site, as the hydrazone is stable at pHs above 7, and can be cleaved under acid pHs. For example, the hydrazone can be used as a release point in a delivery system.. Spacer will reduce or eliminate problems with aggregation and immunogenicity, while adding much needed water solubility -- often huge issues with conventional heterobifunctionals Applications: The general application of this class of heterobifunctional crosslinkers is the controlled and selective conjugation of an amine- containing target (reacts with the NHS ester), with a carbonyl containing complementary target molecule after the t-boc hydrazide has been deprotected with either TFA or HCl. Many biological molecules contain the amine function, or can have it incorporated, and the same can be said for the carbonyl moiety, in small molecule drugs, as well as peptides, oligos or other complementary biologicals. The chief advantage of the dPEG containing heterobifunctional crosslinkers over conventional hydrophobic methylene spacers is that the dPEG spacer is very hydrophilic/water soluble and non-immunogenic spacer. These properties also can work to eliminate aggregation issues often encountered. The extreme hydrophobicity of conventional reagents has made them a challenge to apply, and non-optimal solutions. A new generation of heterobifunctionals is now available. IN ADDITION, conventional linkers have gone to shorter spacers to get around some issues of antigenicity, but one generally needs longer spacers, the dPEG series provides the ideal NEW options in heterobifunctional crosslinking Other spacer lengths will be added to our product selection in the future.
